摘 要:目的 探讨FHIT基因蛋白 (Fhit)表达情况与乳腺癌发生发展的关系。方法 用免疫组化法检测了6 1例乳腺浸润癌 ,4 3例导管内癌及上皮不典型增生 ,37例导管上皮增生和 4 2例正常乳腺上皮Fhit表达水平。结果 90 .5 %正常乳腺上皮Fhit表达为强阳性 ,有 4 9.2 %乳腺癌Fhit表达阴性或明显降低 ,另有 32 .8%中度降低 (P <0 .0 0 1) ;在乳腺上皮增生发展为癌的过程中 ,Fhit表达水平逐渐降低 (P <0 .0 5 ) ;有异常 p5 3蛋白表达的乳腺癌 ,其Fhit低表达率明显高于无异常p5 3蛋白表达者 (P <0 .0 5 )。结论 实验结果提示Fhit表达降低可能对乳腺癌的演化和进展具有一定重要作用 ,Fhit低表达可能与 p5Purpose: To investigate the possible relationship between FHIT protein (Fhit) expression and the breast tumor progression from premalignant in situ to invasive carcinoma. Methods: The expression of Fhit was evaluated by using immunohistochemical methods. Studies were performed in 61 cases of invasive carcinoma, 43 cases of carcinoma in situ and atypical hyperplasia, 37 cases of hyperplasia and 42 cases of normal epithelium. Results: About 90.5% normal ductal and lobular epithelium consistently and strongly expressed Fhit. A complete loss or a significant reduction was observed in 49.2% of breast carcinomas. About 32.8% breast carcinomas showed intermediate reduction of Fhit expression (P < 0.001). A statistically significant, negative correlation in Fhit expression among the stages of disease progression from normal to invasive carcinoma was observed (P < 0.05). Statistically significant differences in the presence of Fhit (-)/p53(+) and Fhit(-)/p53(-) patterns were noted in invasive carcinoma (P < 0.05). Conclusions: The results indicate that the reduced expression of Fhit may play an important role in the development and progression of the breast carcinoma. There may be an association between expression of Fhit and p53 alteration.
