生长激素受体在胰腺癌细胞的表达及生长激素对胰腺癌细胞周期的影响

摘　　要：目的　观察生长激素受体 (GHR)在胰腺癌细胞的表达及生长激素 (GH)对胰腺癌细胞周期的影响。方法　用免疫组织化学方法检测胰腺癌细胞SW 1990中的GHR ;SW 1990中加入GH(5 0、10 0 μg/L)培养 2 4、48、72h后计数细胞 ,2 4h后以流式细胞仪测定细胞周期 ;裸鼠成瘤后分为实验组 (GH组 ,按剂量及时间分为 4mg 2周、2mg 2周、2mg3周 3个亚组 )及对照组 (NS组 ) ,用药期间测体重及瘤大小。结果　GHR在SW 1990上有低度表达 ;加入GH培养 2 4、48、72h后细胞数增加 ,2 4h后细胞S %及增殖指数 (PI)提高 [5 0 μg/L组的S % (5 2 .0 0± 14 .83 )及PI(0 .72±0 .0 9)、10 0 μg/L组的S % (5 3 .2 1± 15 .49)及PI(0 .70± 0 .0 9)均分别高于对照组 (S % :3 0 .15± 6.2 2 ;PI :0 .5 2± 0 .13 )(P <0 .0 5 ) ] ;GH不加速种植瘤生长。结论　少量GHR存在于SW 1990细胞 ,GH在体外能促进SW 1990细胞增殖 ,但在体内不刺激肿瘤生长。ObjectiveTo observe the expression of growth hormone receptor in pancreatic carcinoma,and the effect of recombinant growth hormone on pancreatic carcinoma cell cycle.MethodsThe expression of growth hormone receptor in SW-1990 human pancreatic carcinoma cells was detected by using immunohistochemistry techniques.Cells during exponential growth stage were harvested and cultured in medium containing growth hormone with various concentrations (50 μg/L,100 μg/L).After 24,48,72 h,cells were counted using a Coulter Counter.Cells cultured for 24 h were made into single cell suspentions and samples underwent subsequent cell cycle evaluation.Athymic nude Balb/c mice were injected with SW-1990 cells.When tumor became palpable obviously,animals were randomized to receive r-hGH (Group A,4 mg/kg.s.c.once daily for 2 weeks;Group B,2 mg/kg.s.c.for 2 weeks;Group C,2 mg/kg.s.c.for 3 weeks) versus saline control.Animals were weighed daily and tumor size was measured every other day.ResultsGHR was weakly expressed in SW-1990 in contrast to lymphocytes.GH stimulated cell growth in vitro and also elevated percent S phase (50 μg/L:52.00±14.83 vs 30.15±6.22,and 100 μg/L:53.21±15.49 vs 30.15±6.22,respectively,P <0.05) and proliferation index (50 μg/L:0.72±0.09 vs 0.52±0.13,and 100 μg/L:0.70±0.09 vs 0.52±0.13,respectively,P <0.05) of cells after 24-h culture.GH did not accelerate tumor growth in vivo.ConclusionThere are a small quantity of growth hormone receptors in SW-1990 cells;GH stimulates cell growth in vitro and elevates percent S phase of cells but doesn't accelerate tumor growth in vivo.
