摘  要:目的 观察正常大鼠脊髓中血红素氧和酶 (HO)的表达和分布。方法 雌性SD大鼠10只 ,按体重配成 5对。每对随机分为正常组和诱导组 ,后者予以 42℃热水浸泡躯体 2 0min。取其中 3对大鼠胸腰段脊髓 ,提取RNA后进行北方印迹分析 ;2对做免疫组织化学检测。结果 HO 1探针检测到 1.8kbmRNA ,诱导组的丰度数倍于正常组。HO 2探针检测到两组标本都表达两种同源mRNA(1.3kb和 1.9kb) ,表达量相仿。免疫组织化学显示HO 1阳性细胞主要分布于前角神经元 ,诱导鼠中间带也可出现 ;HO 2阳性细胞散布于整个灰质中。结论 HO参与脊髓信号传导和应激反应。ObjectiveTo investigate the expression of heme oxygenases (HO) in rat spinal cord and its significance.MethodsTen female SD rats were paired and randomly assigned into normal group and induced group.The latter was subjected to 42 ℃ hot water bath for 20 min.Total RNA of the spinal cord was extracted from the first 3 pairs,and the last 2 pairs underwent immunohistochemistry analysis.ResultsHO-1 probes detected 1.8 kb mRNA in all samples,and the messages in heat-induced rats were increased by folds.Blot hybridization with HO-2 probe revealed the presence of two homologous transcripts (1.3 and 1.9 kb) in all samples with relatively equal abundance.Immunohistochemistry revealed the HO-1 mainly distributed in anterior horn,and it was also found in middle region in induced rat.HO-2 positive cells scattered in the whole gray matter.ConclusionHO may involve in message conduction and stress reaction.
