摘  要:目的 探讨外周血淋巴细胞HPRT基因突变作为筛选肿瘤高危人群的一个生物学标志的可能性。方法 采用多核细胞法研究肿瘤患者及其接受放疗后外周血淋巴细胞HPRT基因位点的突变频率 ,并与正常人群进行对比。结果 肿瘤患者的外周血淋巴细胞HPRT基因突变频率均较正常对照组高 ,接受放疗的肿瘤患者外周血淋巴细胞HPRT基因突变频率较放疗前显著升高。结论 HPRT基因位点突变适合于肿瘤高危人群的筛选和生物学检测 ,有望成为癌患风险评价的一个生物学标志。Objective To explore the possibility of HPRT locus mutation in peripheral blood lymphocytes as a biomarker for screening those tumor susceptible people.Methods Multinuclear cell assay was used to investigate the frequency of hprt locus mutation both in normal healthy people and malignant tumor patients prior to and after radiotherapy with a cumulative dose of 54~72Gy.Results The frequency of hprt locus mutation in malignant tumor patients were higher than that in normal healthy people (P<0.05).Compared with malignant tumor patients who didn't receive radiotherapy,the frequency of hprt locus mutation of that received radiotherapy were significantly increased (P<0.001).Conclusions The hprt locus mutation in peripheral blood lymphocytes is useful for screening and bio-monitoring those tumor susceptible people,and may be expected to be a biomarker for cancer risk assessment.
