摘  要:目的 :研究不同氧化相关因素对中国仓鼠肺成纤维细胞 ( CHL )和 He L a细胞 DNA损伤的自身修复情况。方法 :将 CHL细胞和 He La细胞用不同氧化相关因素处理一定时间 [CHL细胞 :过氧化氢 ( H2 O2 ) 2 5 min,重铬酸钾 ( K2 Cr2 O7) 10 5 min,阿霉素 ( Dox) 75 min;He La细胞 :H2 O2 2 5 min,K2 Cr2 O710 5 min],随后立即去毒培养 0、0 .5、1、2、3h,以碱性单细胞凝胶电泳技术检测 DNA链断裂情况。结果 :1CHL细胞经 H2 O2 、K2 Cr2 O7、Dox作用后引起 DNA链断裂 ,去毒培养 1h链断裂修复明显 ( P<0 .0 1) ;去毒培养 2~ 3h,前两毒剂的损伤组完全修复 ,而Dox组链断裂仍高于未损伤组 ;2 He L a细胞经 H2 O2 、K2 Cr2 O7作用后引起 DNA链断裂 ,去毒培养 0 .5 h链断裂明显修复 ( P<0 .0 1) ,去毒培养 1h则完全修复 ;3CHL细胞和 He L a细胞损伤后修复的拖尾率与修复时间的回归系数显著不同 ( P<0 .0 5 )。结论 :两种细胞在氧化性 DNA损伤后均迅速启动自身修复 ,但 He L a细胞比 CHL细胞有更快的修复能力 ;同时这两种细胞由 Dox所致损伤修复能力均较 H2 O2 、K2 Cr2 O7所致的差。Objective: To investigate DNA repair in CHL cells and HeLa cells after DNA damage induced by different oxidative agents. Methods: CHL cells and HeLa cells were exposed to various damaging agents, CHL cells: H 2O 2 for 25 min, K 2Cr 2O 7 for 105 min, doxorubicin (Dox) for 75 min;HeLa cells: H 2O 2 for 25 min, K 2Cr 2O 7 for 105 min; then cells were continuously cultured for 0~3 h after washing. Alkaline single cell gel electrophoresis(ASCGE) assay was used to detect DNA strand breaks. Results: ① DNA strand breaks were induced in CHL cells after exposure to H 2O 2,K 2Cr 2O 7 or Dox, which were repaired evidently after continuous culture for 1 h( P <0.01). The damages induced by H 2O 2 or K 2Cr 2O 7 were repaired completely after culture for 2~3 h. However,the demage induced by Dox was repaired incompletely. ② DNA strand breaks were induced also in HeLa cells after exposure to H 2O 2 or K 2Cr 2O 7, which were repaired evidently after continuous culture for 0.5 h( P <0.01),and completely after culture for 1 h. ③ The regression coefficient related to the rate of comet cells and repair time was statistically different( P <0.05) between CHL cells and HeLa cells. Conclusion: DNA damage induced by Dox is repaired more difficult than that induced by H 2O 2 or K 2Cr 2O 7. The repair initiates immediately after DNA damage in both of cells, but more rapidly in HeLa cells than in CHL cells.
