摘 要:目的 :探讨在高糖环境下肾小球系膜细胞与内皮细胞相互作用对活性氧产生的影响和茶多酚的干预作用。方法 :系膜细胞和内皮细胞单独分别培养和共同培养 ,分正常对照组、高糖组及茶多酚干预组 ,培养 0、12、3 6h后 ,用分光光度法检测培养液中丙二醛 (MDA)含量及超氧化物歧化酶 (SOD)活性。结果 :高糖共同培养时SOD活性比两者分别单独培养时更低(P <0 .0 1) ;共同培养时茶多酚干预组的SOD活性显著高于高糖组 (P <0 .0 1)。高糖共同培养时MDA含量较两种细胞单独培养明显增高 ,高于两种细胞单独培养的总和 (P <0 .0 1) ;共同培养时茶多酚干预组MDA含量低于高糖组 (P <0 .0 1)。结论 :( 1)高糖环境下 ,系膜细胞和内皮细胞存在相互作用 ,这种作用能促进肾细胞活性氧的产生 ;( 2 )Objective To study the effect of high glucose and tea polyphenols(TPs) on the role of oxidative stress via the interaction of mesangial cells and endothelial cells.Methods The model of intercellular interaction between mesangial cells and endothelial cells of diabetic nephropathy was established and divided into 3 groups: control,high glucose and intervented with TPs.The mesangial cells and endothelial cells were co-cultured in RPMI?1640 with or without TPs in high glucose media up to 36 hours.The activity of SOD and the level of MDA were determined by spectrophotometry in the culture media.Results The activity of SOD of co-culture was markedly lower than that from the ordinary monolayer cell culture (P<0.01 ).The activity of SOD of co-culture intervented by TPs was much higher than that of high glucose group(P<0.01).The level of MDA of co-culture was higher than that of the ordinary monolayer cell culture(P<0.01).The level of MDA of the group intervented by TPs was lower than that of high glucose group(P<0.01).Conclusions (1) Exposed to high glucose,mesangial cells and endothelial cells can interact with each other to promote ROS,which would break the balance of oxidant state and accelerate the development of diabetic nephropathy.(2)The intercellular interaction between mesangial cells and endothelial cells may be intervened by TPs,which are able to eliminate the excess of ROS.
