摘  要:利用自行筛选的Pseudomonassp.js-01菌发酵产酶,得到兼有海因酶、N-氨甲酰水解酶活性的菌体,通过酶动力学参数分析以及转化试验表明,N-氨甲酰水解酶活性偏低是制约整体转化过程的瓶颈。以疏水层析的方法将海因酶、N-氨甲酰水解酶进行初步分离,并调节双酶比例进行了转化试验。结果表明,转化过程得到显著改善,当海因酶与N-氨甲酰水解酶比例达到1∶4时,中间产物的积累基本消失,6h产物转化率提高1倍以上。The free cells containing D-HEn and N-CAEn activity were obtained by fermentation using Pseudomonas sp.js-01 screened.It is demonstrated that the low activity of N-CAEn was the limiting factor of the whole conversion process by the dynamic parameters determination and conversion experiment.D-HEn and N-CAEn were separated using HIC,and the ratio of the two enzymes was adjusted and the experiment results show that the conversion process is promoted sharply.The conversion ratio of D-Phe with D-HEn\\N-CAEn as 1∶4 within 6 h is two times more than that as 1∶1.
