摘  要:目的 :探讨容量调控氯通道在人胃癌细胞增殖中的作用 .方法 :应用细胞计数及MTT比色分析法观察细胞增殖情况 ;用流式细胞仪检测细胞周期时相 ;用膜片钳技术观察NPPB对容量调控氯通道的作用 .结果 :NPPB (1 0 0 μmol/L)能够可逆性地抑制SGC790 1细胞数及MTT吸光度值 .经NPPB处理 4 8h后 ,G1 细胞比例比对照组明显增高 .给予NPPB后可以抑制低渗激活的氯电流 .结论 :容量调控氯通道在人胃癌细胞系SGC790 1的增殖中发挥重要作用 .AIM: To study the role of volume regulated chloride channel in the proliferation of human gastric carcinoma cells. METHODS: Cell proliferation was analyzed by cell counting and MTT assay and the cell cycle analysis was carried out by flow cytometry. The currents of volume regulated chloride channel were recorded by patch clamp. RESULTS: NPPB (100 μmol/L), a chloride channel blocker, reversibly decreased the cell number and the MTT optical density (OD) of SGC7901. Compared with control, NPPB arrested cells in G 1 phase and extracelluar application of NPPB inhibited volume regulated chloride currents which were induced by hypotonic stress. CONCLUSION: The volume regulated chloride channel plays an important role in the proliferation of gastric carcinoma cell.
