摘 要:目的 研究重组腺相关病毒介导的血管内皮生长因子 165反义RNA(rAAN_aVEGF165)抑制碱烧伤后角膜新生血管的作用。方法 3 0只SD大鼠随机分成 2组 ,每组 15只。实验组前房注射rAAV_aVEGF165,对照组前房注射rAAV_Lacz。 3 0天后 ,碱烧伤诱导角膜新生血管 ,以形态学分析评价角膜新生血管的情况 ,Westernblot评价角膜VEGF的表达。结果 形态学分析显示实验组角膜新生血管得到抑制 ,Westernblot结果显示 ,碱烧伤后各个时期角膜VEGF165的表达实验组明显低于对照组。结论 下调VEGF能有效抑制角膜新生血管的形成 ,重组腺相关病毒是眼反义基因治疗的有效载体。Objective This study was to determine whether rAAV_mediated gene transfer of antisense vascular endothelial growth factor 165 (aVEGF 165) mRNA inhibits the development of corneal neovascularization (CNV) in a rat model. Methods The ocular anterior chamber of spragud-Dawleg (SD) rats were injected with rAAV-aVEGF 165(10 10 pfu), another group of SD rats were injected with rAAV-Lacz (10 9 pfu) in anterior chamber as control. CNV in rat was induced by alkaline cauterization of the central cornea,30 days after the viral vector injection. The occurrence and development of CNV was observed by slit-lamp microscope,and the area of CNV was calculated. Experssion of VEGF 165 was evaluated by Western blot.Results Morphometric analysis demonstrated rAAV-mediated antisense VEGF 165 significantly inhibits corneal neovascu larization.The VEGF 165 protein were decreased at 1、2、4、30 days postcautery in treatment groups compared to control group.Conclusion This study suggest that the specific down_regulation of vascular endothelial growth factor production is sufficient to reduce the corneal neovascular response.
