摘　　要：目的 :研究阳离子脂质体Lipofectin介导的 β 半乳糖苷酶基因在不同肿瘤细胞株中的表达。 方法 :质粒pDCβ在大肠杆菌DH5α中扩增后用碱裂解法提取 ,并经Sepharose 2B凝胶过滤柱层析 ,以Lipofectin分别转染人肝癌细胞株HepG2 、人肾癌细胞株GRC ,然后用X gal细胞化学染色法检测 β 半乳糖苷酶活性。 结果 :β 半乳糖苷酶活性在HepG2 、GRC中均有表达。结论Aim: To investigate the transfection and expression of β galactosidase reporter gene mediated by cationic liposome in 2 tumor cell lines. Methods: Plasmid pDCβ were amplified in E.coli DH 5α strain cells, extracted by alkaline lysis and purified by passage through Sepharose 2B column chromatography. 2 cell lines including human hepatocarcinoma cell line HepG 2, human renal carcinoma cell line GRC were used to be transfected by cationic liposome Lipofectin. The activity of β galactosidase gene were detected by X gal chemical stain. Results: The activity of β galactosidase gene were detected in 2 cell lines. Conclusion: Cationic liposome Lipofectin is a efficient cytofectin capable of transferring DNA into different tumor cells.
