摘　　要：目的 :通过研究结核特异性转移因子 (transferfectortuberculosis,TFtb)对巨噬细胞促进BCG预致敏淋巴细胞增殖反应的影响 ,探讨转移因子 (TF)在传递特异性抗原信息过程中的作用机制。方法 :将小鼠腹腔巨噬细胞作为刺激细胞 ,脾脏非贴壁细胞为效应细胞 ,PPD作为抗原 ,以不同浓度的TFtb为刺激剂 ,用MTT法检测T细胞的增殖率。结果 :预致敏鼠的淋巴细胞增殖率随TFtb浓度升高而增高 ,0 .5mg·ml-1 时达到峰值 ,随后浓度升高增殖率反而下降。TFtb促预致敏淋巴细胞增殖是巨噬细胞依赖性和抗原特异性的。对未致敏鼠的淋巴细胞增殖率无明显改变。结论 :TFtb能够增强巨噬细胞促结核抗原预致敏淋巴细胞增殖反应的作用 ,可能与增强巨噬细胞呈递抗原的作用有关 。Objective: To study the effect of TFtb on proliferation of primed lymphocyte induced by macrophages and to explore the mechanism of TF on the procession of transfer antigen special message. Methods: Murine macrophages were cultured together with PPD and treated with different concentration TFtb. The effects of macrophages on the lymphocyte were detected by MTT colorimetric assay. Results: The higher of the concentration of TFtb, the higher of the lymphocyte proliferation rate. When it was up to 0.5 mg·mL -1 , the rate of proliferation achieved peak. Then the rate was descent when the concentration was beyond that. The proliferation of primed lymphocyte was depended on macrophage and special antigen. While there was no effect on the lymphocyte cells from unprimed mice. Conclusion: The effect of TFtb on proliferation of primed lymphocyte induced by macrophages may have relationship with the procession of antigen presenting by macrophages. And the effect of TF is concentration dependence.
