摘  要:目的:探索叶酸缺乏状态对人肝癌细胞株BEL7402、肝细胞株L02生长状况的影响。方法:将BEL7402、L02分别置于正常培养液和叶酸缺乏培养液中培养30d,利用细胞计数和流式细胞仪检测各组细胞增殖和凋亡状态。结果:L02在叶酸缺乏状态下凋亡率增加,生长旺盛,表现为细胞总数增多,G0、G1期细胞比例下降。BEL7402在叶酸缺乏状态下生长正常,细胞总数、细胞周期比例和凋亡率未受影响。结论:叶酸缺乏状态刺激人肝细胞株L02生长旺盛、凋亡率增加,而对人肝癌细胞株BEL7402则作用不明显。叶酸缺乏可使L02细胞凋亡率明显增加,若凋亡机制未能清除那些不发生G1期阻滞的细胞,则可能发生癌变。Objective To study the effect of folate deficiency status on the growth and apoptosis of human liver cell strain L,02 and human liver cancer strain BEL,7402. Methods L,02 and BEL,7402 were cultured in the normal medium and the medium which contain less folate for 30 days. The proliferation and apoptosis of these cells in the two different culture media was examined by cell counter and flow cytometry(FCM). Results In the medium which contain less folate, the ratio of apoptosis was increased, the growth of L,02 were stimulated ,manifesting the percentage of G0G1 cells decreased.However,human cancer cell strain BEL,7402 grew as quickly as in the normal medium. Conclusion The results of folate deficiency status on the growth and apoptosis of L,02 and huaman liver cancer strain are different.Folate deficiency makes the apoptosis ratio of L,02 increase, if the mechanism of apoptosis can not eliminate the cell which not be inhibited in the phase of G1, the carcinogenesis occurs.
