抗猪脂肪细胞膜蛋白抗体的制备与鉴定

摘　　要：以蔗糖密度梯度离心提取猪皮下脂肪细胞和其他组织细胞膜蛋白,经SDS-PAGE分析。Membrane proteins of pig subcutaneous adipocyte and other tissue cells were extracted by sucrose density gradient centrifugation. Though components of membrane proteins were different among pig subcutaneous adipocyte and other tissue cells, some membrane proteins were identical. The antibody against adipocyte membrane proteins were produced by immunization with subcutaneous adipocyte membrane proteins as antigen, and its titer was 1∶12 800 detected by ELISA. Western blotting showed that the antibody reacted with membrane proteins in cells of heart, hepatocyte, spleen, kidney, lung, skeletal muscle, erythrocyte, small intestine fat and fat surrounding kidney, but the reaction ability was weaker. The results demonstrated that the antibody has specialty to subcutaneous adipocyte and higher titer.
