摘 要:背景:研究发现晶体上皮细胞的凋亡是白内障发生的共同细胞学基础。目的:观察阿司匹林对平阳霉素诱导培养兔晶体白内障形成的抑制作用,研究阿司匹林对晶体上皮细胞凋亡的抑制作用。设计:随机对照实验设计。地点和对象:实验在江西医学院第二附属医院眼科完成,对象为健康家兔,1.5~2.5kg,雌雄不拘,由江西医学院动物室提供。干预:采用兔晶体体外培养方法,观察阿司匹林对不同培养时间(24,48,72,96h)平阳霉素诱导兔晶体混浊形成的影响。在透射电镜下观察晶体上皮细胞超微结构的改变。通过免疫组织化学S-P染色检测不同培养时间(12,24,36h)晶体上皮细胞Bcl-2表达的情况。主要观察指标:晶体混浊度、上皮细胞超微结构和Bcl-2表达。结果:阿司匹林可延缓和减轻平阳霉素诱导的兔晶体混浊的形成。培养24h透射电镜下平阳霉素组和平阳霉素+阿司匹林组均检测到凋亡细胞。阿司匹林+平阳霉素组和平阳霉素组Bcl-2染色的积分吸光度在12h分别为251.01±75.27和122.23±40.74(P<0.05),在24h分别为178.63±16.84和96.15±15.76(P<0.05),在36h分别为114.14±29.86和64.71±15.53(P<0.05)。结论:阿司匹林可抑制平阳霉素诱发的兔晶体上皮细胞凋亡,延缓和减轻平阳霉素诱导的兔白内障的发生和发展。BACKGROUND:It was reported that the apoptosis of lens epithelia was the common cytological basis of the genesis of cataract. OBJECTIVE:To observe the inhibitory reaction of Aspirin on the genesis of cataract in lens of rabbits induced by Pingyangmycin and to study the inhibitory reaction of Aspirin on the apoptosis of lens epithelia as well. DESIGN:A randomized controlled study was performed. SETTING and PARTICIPANTS:This study was completed in the Department of Ophthalmology,the Second Affiliated Hospital of Jiangxi Medical College.The subjects were healthy rabbits in either gender with a body mass from 1.5 kg to 2.5 kg obtained from Laboratory for Animals,Jiangxi Medical University. INTERVENTIONS:The impact of Aspirin on the genesis of the turbidity in lens of rabbits induced by Pingyangmycin over different period of culture(24,48,72 and 96 hours) by the method of rabbit lens culture in vitro.The changes of ultrastructure of lens epithelia were observed under transmission electron microscope.The expression of Bcl 2 in lens epithelia over different culture period(12,24 and 36 hours) was observed through immunohistochemical S P staining. MAIN OUTCOME MEASURES:The turbidity of lens,the ultrastructure of epithelia,and the expression of Bcl 2. RESULTS:Aspirin could delay and alleviate the genesis of turbidity in lens of rabbits induced by Pingyangmycin.Apoptosis cells were detected at 24 hours after culture under transmission electron microscope in both Pingyangmycin group and Pingyangmycin+Aspirin group.The integral absorbance(A) of Bcl 2 staining at 12 hours after culture in Aspirin+Pingyangmycin group and Pingyangmycin were 251.01±75.27,and 122.23±40.74 respectively(P< 0.05);and at 24 hours after culture were 178.63±16.84 and 96.15±15.76 respectively(P< 0.05);and at 36 hours after culture were 114.14±29.86 and 64.71±15.53 respectively(P< 0.05). CONCLUSION:Aspirin can inhibit the apoptosis of lens epithelia in rabbits induced by Pingyangmycin and therefore,it can delay and alleviate the genesis an
