白血病患者外周血细胞Cu、Zn-SOD含量变化的观察

摘  要:本文报告50例健康成人。32例白血病患者血RBC、LC及GC、Cu、Zn-SOD放免分析结果。健康成人血LC、Cu、Zn-SOD含量明显>GC,此与文献报道正常人LC、Cu、Zn-SOD活性>GC相一致。32例白血病患者血RBC、Cu、Zn-SOD均明显降低(P<0.05),反映RBC清除胞内O(?)和维护胞膜结构完整性的功能减弱。其中53.13%白血病患者LC、Cu、Zn-SOD呈非常显著性增高(P<0.002),56.25%病人血LC、Cu、Zn-SOD水平未见明显升高。本文结果揭示淋、粒细胞白血病血细胞中Cu、Zn-SOD出现异常改变,O(?)随白血病细胞反应而产生增多,导致血细胞内Cu、Zn-SOD与O(?)平衡失调,从而自由基可损伤SOD基因表达和调节功能,表明Cu、Zn-SOD,O(?)与白血病的发生,发展有一定的内在联系。预测在抗白血病治疗中辅以SOD治疗,则将有助于控制血液肿瘤的发展。This report describes a Radioimmunoassay for the quantitation of Cu、Zn superoxide dismutase in erythrocytes, granulocytes and lymphocytes of 32 patients with leukemia and 50 Healthy subjects. A content of Cu、Zn SOD in normal lymphocytes is greater than that in granulocytes. This result is in agreement with the Cu、Zn SOD activity in lymphocytes from human peripheral blood which is considerably higher than that in polymorphonuclear leucocytes found by other reports. In the group of 32 paticnts with leukemia, the Cu、Zn SOD Ievel in red blood cells are markedly decreased (P<0.05), which reflect that the function of scavenging the O_2^(?)in RBC and protecting the construction integrity of RBC membrane was diminished. The Cu, Zn SOD in lymphocytes was found to be significantly increased (P<0.002) in 53.13% of patients with leukemia. But 56.25% of patients with leukemia arc not significant changes of Cu、Zn SOD level in granulocytes, as compared to control group. The data reveals that an abnormality of the Cu、Zn SOD in leukemic blood cells. and the supcroxide anion (O_2.(?)) produced increased by the reaction of leukemic cells, followed by the disorder of a balance between the leukmic intracellular Cu、Zn SOD and O_2^(?)was produced. Therefore, regulating function and the expression of SOD gene was impaired by free radicals, which suggest the Cu、Zn SOD and O_2^(?) is clossly linked with the generation and development of leukemias. We assume that a assistant treatment with Cu、Zn SOD in anti-leukemic therapy, which is an aid to controlling the development of maligant blood tumors.
