摘  要:目的:研究丹参单体IH764-3在大鼠体内、外的抗肝纤维化作用,重点观察对胶原降解的影响。方法:40只SD大鼠随机分为3组,对照组10只,模型组及IH764-3组各15只。以胆总管结扎法(BDL)建立肝纤维化模型,用免疫组化法检测大鼠组织血细胞金属蛋白酶组织抑制因子(TIMP-1)、转化生长因子β_1(TGF-β_1)和肝组织羟脯氨酸(Hyp)含量。结果:BDL结扎14d后有明显肝纤维化形成。IH764-3使肝组织Hyp减少40.6%,同时TIMP-1、TGF-β_1蛋白质表达分别下降36.2%和32.5%;在体外使TIMP-1、TGF-β_1阳性肝星状细胞数目减少。结论:丹参单体IH764-3具有抗肝纤维化作用,其机制之一是抑制TIMP-1和TGF-β_1蛋白的表达。To observe the role of salvia miltiorrhiza monomer IH764-3 in anti-fibrogenesis in vivo and in vitro, especially the mechanism involved in collagen degradation. Methods: Forty male SD rats were divided into three groups at random, namely control group(n=10), model group (n=15) and IH764-3 group(n=15). The experimental hepatic fibrosis was induced by common bile duct ligation(BDL). The tissue sections and the cover slides of hepatic stellate cells(HSC) were stained with immunohistochemistry to detect tissue inhibitor of matrix metalloproteinase-1(TIMP-1), transforming growth factor beta-1 (TGF-β_1) and hydroxyproline(Hyp) positive areas of tissue sections were measured. Results: Obvious hepatic fibrosis had come into being 14 days after BDL. IH764-3 decreased the content of Hyp by 40.6% and the positive area of TIMP-1 and TGF-β_1 by 36.2% and 32.5% respectively. In groups of HSCs exposed to different dose of IH764-3, the numbers of positive cells of TIMP-1 and TGF-β_1 decreased evidently. Conclusion: Silvia mitiorrhiza monomer IH764-3 may relieve hepatic fibrosis in vivo and in vitro, one of the mechanisms of which seems to be relative to decreasing expression of TIMP-1 through inhibiting expression of TGF-β_1.
