摘 要:目的 探讨乙肝散逆转实验性肝纤维化过程中α -SMA阳性HSC变化。方法 雄性Wistar大鼠尾静脉注射白蛋白建立免疫损伤性肝纤维化模型后 ,分组加用不同浓度的乙肝散颗粒饲料喂养 12周 ,流式细胞仪定量分析大鼠肝组织α SMA阳性肝星状细胞 (HSC) ,观察大鼠肝组织病理、肝组织Lillie和苦味酸 天狼红染色、Ⅳ胶原免疫组化、α SMA免疫组化、肝匀浆羟脯氨酸。结果 乙肝散应用组肝组织α SMA阳性HSC较模型对照组明显降低 (P <0 0 1) ,同时肝组织Ⅰ胶原、Ⅲ胶原、Ⅳ胶原水平和Hyp含量明显降低 (P <0 0 1或P <0 0 5 )。结论 α SMA阳性HSC减少是实验性肝纤维化逆转过程中重要的因素 ,乙肝散促进活化HSC减少。? Objective To explore thechange of activated hepaticstellate cellsin the process of reversing immuno-damaged hepatic fibrosisby Yigansan. Method The dilution albumin was injected into the caudal vein of selected male Wistar rats to establish the experimental model of immuno-damaged hepatic fibrosis. Then the rats were fed with different concentrations of Yigansan particles in different groups respectively for 12 weeks. Theactivated hepaticstellate cellsin liver tissues were quantitatively analyzed by flow cytometry (FCM), while liver homogenate hydroproline content, α-SMA, and collagen Ⅰ, Ⅲ and Ⅳwere determined by biochemical and immunohistochemical methods respectively. Result Compared with that in the model group, thenumber of activated hepaticstellate cellswassignificantly decreased (P<0.01), and the levels of collagen Ⅰ, Ⅲ, Ⅳ as well as Hyp in the liver tissue were significantly lower(P<0.05 or P<0.01)in the treated groups.Conclusion Thedecrease of activated hepatic stellate cells in the liver tissue plays an important parts in the process of reversing immuno-damaged hepatic fibrosis.Yigansan has the function of reducing activated hepaticstellate cells.
