草原兔尾鼠卵透明带3在Pichia pastoris酵母中的分泌表达

摘　　要：本研究通过酵母Pichiapastoris系统表达草原兔尾鼠卵透明带 3(lZP3)蛋白 ,获得控制害鼠生育研究的抗原物质。根据本实验室克隆获得的lZP3基因序列设计引物 ,并分别在 5’引物和 3’引物中引入了EcoRI和XbaI酶切位点。经PCR扩增 ,将lZP3克隆至穿梭质粒pSuperY上 ,获得的重组穿梭质粒pSuperY -lZP3经线性化后 ,采用电激法将重组穿梭质粒转入酵母SMD116 8菌株中 ,Zeocin+筛选阳性克隆 ,经小瓶发酵后 ,取上清作SDS -PAGE和Western -blot检测 ,结果表明lZP3在酵母中得到了成功表达 ,表达产物的分子量约为 5 5kD 。Formation of the egg's extracellular matrix,the zona pellucida3,is critical for fertilization.The purpose is to express Lagurus lagurus zona pellucida3 ( ZP3 ) in Pichia pastoris.A 1065bp DNA fragment without the N-terminal signal peptide and C-terminal transmembrane domain was inserted into the downstream of the a-mating factor secretion signal in the expression vector pSuper Y.The construct was linearized and introduced into the host SMD1168 genome by the electroporation under the selection of zeocin. Then the positive colonies were small-scaled in 20ml YPD.SDS-PAGE and Western-blot analysis indicated that the expression product was about 55kD.
