摘 要:利用紫外分光光度法建立一种快速检测和定量测定灵芝中三萜类化合物含量的方法.以饱和NaHCO3溶液溶解灵芝三萜单体化合物标准品后,采用紫外分光光度法测定溶液在257nm处具有最大吸光度,标准品在0~200.0μg·mL-1范围内,吸光度与浓度呈良好的线性关系,相关系数r为0.99988.利用紫外分光光度法在提取三萜类化合物工艺的中间阶段,即碱提的NaHCO3层进行检测,根据标准曲线可计算出样品中三萜类化合物的含量.该测定方法简单快速、准确度高、实验误差小、重复性好,可以应用于灵芝及其相关产品的质量评估.A method for the rapid quantification of triterpenoids in Ganoderma lucidum by ultraviolet (UV) spectrometry is established. A triterpenoid standard dissolved in saturated NaHCO_3 has a maximum absorbance at 257 nm by UV spectrometry. The concentration of the standard is correlated very well to its optical density at 257 nm in the range of 0~(200.0 μg·mL^(-1)) with a relativity coefficient of (0.999 88.) The supernatant NaHCO_3 solution was collected and measured in the intermediate stage of triterpenoids extraction by a UV spectrometer. The content of triterpenoids in Ganoderma lucidum material was then counted according to the standard curve. The above method for the quantification was proved to have the advantages of easy operation, high accuracy, having less error and fine repetition, and it shound be helpful for monitoring the quality of Ganoderma lucidum material and related products.
