天然无机骨材料肌内短期植入后的组织学和酶学观察

摘  要:目的 :采用肌内植入模型 ,对猪股骨去抗原性来源的天然无机骨 (NaturalInorganicBone ,NIB)生物相容性进行评价 ,为临床骨缺损修复材料的选择提供依据。方法 :采用有毒材料 (Organo -pbstabilizedPolyvinyl chioride,OP)作为阳性对照 ,并设单纯创口组 (Open -Close ,OC)排除手术影响 ,运用一般组织学和细胞酶组织化学方法动态观测肌内植入模型中组织对材料的反应及有关酶活性的变化。结果 :NIB材料初始可引起周围组织以中性粒细胞浸润为主的炎性反应 ,至 15天炎症反应渐消退 ,30天时材料周围有薄层包膜形成 ,肌肉组织形态、排列恢复正常 ;OC组在 10天时炎症反应消退 ,30天时瘢痕修复创口 ;OP植入体周肌肉组织炎症反应在观察期内未消退且向更大范围波及。酶组织化学结果显示 :NIB植入体周围肌肉组织内ACP活性早期升高显著 ,而其他标志酶活性在早期有所下降 ,30天所有酶学指标恢复正常 :OC组酶组化结果与NIB材料组类似 ;OP材料周肌组织内各种酶活性在第 30天时仍偏离正常范围。结论 :NIB材料具有良好的生物相容性。Objective:To evaluate biocompatibility of natural inorganic bone(NIB)harvested from the xenogeneic bone,and provide further experiment evidences for the choice of materials in plastic surgery.Methods:NIB were obtained from pig femur treated with physical and chemical methods.On the basis of histological evaluation,enzyme analysis was used for further understanding of NIB whether the cells surrounding these materials were injured or not.Toxic material Organo-pb stabilized Polyvinylchioride(OP)were employed as positive control materials,and the open-close group(OC)was set to rule out the affection of surgical procedure.Results:Acute inflammatory response of tissue was observed at the early period on NIB group.Neutrophils disappeared at 15 days after operation.Fibrous membrane formation around NIB cylinders,and shape and array of muscle fibrous recovered at 30-day after operation;Whereas inflammatory response to OP was still observed at 30 days after implantation.Nutrophils immersion was disappeared at 10 days after operation on OC group,and the wound of muscle tissue was repaired by scar tissue formation at the 30th day;On the other hand,the enzyme analysis showed that the activity of ACP drastically increased at early period then decrease slowly,and the activity of other indicative enzymes exhibited the reverse behave on NIB group.All of these enzymes activity recovered at the end of the observation period;The six kinds of enzymes showed the similarity of changes on OC group;Whereas these enzymes in tissue around OP implanters had not recovered in the same period.Conclusion:NIB material has good biocompatibility.
