阿尔茨海默病和血管性痴呆患者血管紧张素转换酶和载脂蛋白E基因多态性分析

摘　　要：目的　探讨阿尔茨海默病 (AD)及血管性痴呆 (VaD)与血管紧张素转换酶 (ACE)和载脂蛋白E(apoE)基因多态性的关系。方法　应用聚合酶链反应和限制性片段长度多态性方法 ,检测了 2 6例晚发AD患者、5 4例VaD患者和6 8例正常老年人的ACE和apoE基因多态性。结果　AD组中apoE等位基因频率分别为ε2 0 .0 77、ε30 .6 15及ε40 .30 8,VaD组apoE等位基因频率分别为ε2 0 .0 5 6、ε30 .6 85及ε4 0 .2 5 9,AD组和VaD组apoEε4等位基因频率显著高于对照组。ACE基因AD组和VaD组DD型频率高于对照组 ,D等位基因亦高于对照组。结论　ACEDD型。Objective To explore the relations between the polymorphisms of apolipoprotein E(apoE) gene and ACE gene and Alzheimer's disease(AD) and vascular dementia(VaD).Methods The polymorphisms of ACE gene and apoE gene were detected by polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) technique in 26 patients with late-onset AD,54 patients with VaD and 68 normal controls. Results The frequencies of allele ε2,ε3 and ε4 of apoE gene were 0.077,0.615 and 0.308 respectively in AD patients and 0.056,0.685 and 0.259 respectively in VaD patients. The frequency of ACE genotype DD increased significantly in patients with AD and VaD as compared with the controls.The frequency of allele D increased significantly in patients with AD as compared with the normal control.Conclusions The results indicated that ACE genotype DD and allele ε4 of apoE gene might increase the risk of the AD and VaD.It is significant to monitor and intervene the risk factors of dementia for decreasing the incidence.
