摘  要:目的 研究抗脑抗体———神经突触前膜胞内蛋白 (Munc 18)抗体是否能慢性点燃致痫大鼠以及其致痫的机制。方法  在SD大鼠的海马CA1区注射Munc 18抗体 ,隔天注射一次 ,5次之后每隔两周注射一次 ,共 10周。期间监测大鼠的脑电图 (EEG)和痫样行为。 10周后取脑切片作HE、尼氏和TUNEL染色。 结果  实验组12只大鼠中 ,有 10只有痫样EEG(83% ) ,5只 (5 0 % )在 6周之后仍有保留 ;9只有 14级痫样行为 (75 % ) ,6周以后为 4例 (4 4% ) ;染色发现在海马区有胶质细胞增生 ,神经元细胞数量减少且形态异常 ,凋亡细胞数量明显增多。与对照组相比均有显著差异 (P <0 .0 1或P <0 .0 5 )。 结论  Munc 18抗体能慢性点燃致痫大鼠 ,其机制可能与海马区神经元细胞的凋亡有关 ,但还需进一步的研究证实。Objective To investigate whether anti-brain antibody—presynaptic protein ( Munc-18) antibody could induce epilepsy in rat by chronic kindling way and its mechanism. Methods Munc-18 antibody was micro-injected into CA1 area of hippocampus in Sprague-Dawley(SD) rat every other day for five times, then once every other week for additional five times. During injection period, both eletroencepalograph (EEG)and epileptic behaviors were monitored. HE, Nissle and TUNEL staining were applied on rat brain sections after ten weeks. Results Ten of 12 rats (83%)produced epileptiform EEG, 5 cases (50%) among which maintained abnormal EEG after 6 weeks. Nine of the above 12 rats(75%)showed epileptiform behaviors of Racine grade 1 to 4, 4 cases (44%) among which had that after 6 weeks. In hippocampus, neurons showed less numbers and morphologic abnormality, while glial cells proliferated. TUNEL-positive cells significantly increased. Conclusion Munc-18 antibody could induce epilepsy by chronic kindling way which mechanism might be linked to apoptosis in area of hippocampus .However, it needs to be further investigated.
