摘 要:从转化培养基、接种量、底物添加时间、底物初始浓度和底物添加方式5个方面研究了菌株Streptomycessp.108转化美伐他汀(底物)的能力,并在2.5L发酵罐上探索了最适的底物运行浓度。结果表明,适量增加底物浓度,尤其是间歇性添加底物,能够提高普伐他汀产量;当底物运行浓度在450mg/L左右时,转化率可达55%,普伐他汀终浓度可达1088mg/L。The factors such as transformation mediums, the volume of inoculation, the time of adding substrate, the initial concentration of substrate and the ways of adding substrate were investigated in flasks to test the conversion ability of Streptomyces sp.108, which can convert mevastatin to pravastatin, and three running concentrations of substrate were studied in 2.5 L fermentation jar. The results suggested that increasing mevastatin concentration resulted in a higher final pravastatin concentration especially when mevastatin was added intermittently, and that conversion rate was about 55% of mevastatin consumed and the final pravastatin concentration was about 1 088 mg/L when the running concentration of mevastatin was maintained at about 450 mg/L.
