摘 要:目的 :探讨高血压病血管平滑肌细胞凋亡相关基因的表达情况。方法 :Wistar大鼠两肾一夹型高血压模型 ,4周后处死 ,采用免疫组织化学方法 ,观察肾细小动脉平滑肌细胞中Bcl- 6和Bax表达。结果 :实验期间 ,高血压组动脉血压从14 9± 2 4kPa(112± 18mmHg)升高到实验结束时的 2 6 4± 4 4kPa(198± 33mmHg) ;高血压组肾脏小叶间动脉Bcl - 6染色阳性率 (12 96 % )明显高于对照组 (5 2 1% ,P <0 0 1) ;入球动脉、小叶间动脉血管平滑肌细胞 (VSMC)中Bcl - 6阳性率(1 70 %和 5 2 2 % )均高于对照组 (P <0 0 5 ) ;Bax基因蛋白在高血压组肾脏弓形动脉及其VSMC染色阳性率 (分别为1 90 %和 0 90 % )低于对照组 (11 4 3%和 1 6 0 % ) ,P <0 0 5 ;高血压组叶间动脉VSMC中Bax染色阳性率 (2 99%和1 18% )均明显低于对照组 (P <0 0 1) ;在高血压组入球动脉中 ,Bcl- 6和Bax基因蛋白染色之间存在相关性 (χ2 =4 0 0 ,P<0 0 5 ) ;肾细动脉Bcl- 6基因蛋白染色阳性率 (3 0 2 % )明显低于小动脉 (13 6 0 % ) ,P <0 0 1。结论 :两肾一夹型高血压时 ,肾细、小动脉血管平滑肌细胞凋亡相关基因Bcl- 6 /Bax比值升高 ,使得凋亡减少 。Objective: To detect the expression of apoptosis-related genes in vascular smooth muscle cell (BSMC) of hypertension rats. Methods: Two kidney-one clip hypertension Wistar rats were executed and right kidney taken out for histopathological studies immediately after 4 weeks experiment. The expression of Bcl-6 and Bax was observed by means of immunohistochemical technique using respective ELISA kits. Results: During the experiment, blood pressure was raised significantly in the hypertension rats from 14.9±2.4kPa(112±18mmHg) at the beginning to 26.4±4.4kPa (198±33mmHg) in the end. The positive rate of Bcl-6 in hypertensive rat's interlobular artery (12.96%) is significantly higher than that (5.21%)of the controls (P<0.01). The positive staining in VSMC of afferent glomerular and interlobular arteries of the hypertension rats was 1.70% and 5.22% respectively, significantly higher than that of the controls (P<0.05). The positive staining of Bax in arch artery and VSMC of hypertension rats was 1.90% and 0.90% respectively, significantly lower than that of the controls (P<0.05), the positive rate of Bax in interlobar arteries and its VSMC of hypertension rats was 2.99% and 1.18% respectively ,significantly lower than that of the controls (P<0.01). A inverse relationship between the expression of Bcl-6 and Bax was found in afferent arteries (χ 2=4.00, P<0.05). The positie rate of Bcl-6 in arterioles (3.02%) was significantly lower than that in small arteries (13.60%). Conclusion: In two kidney-one clip hypertensive rats, the ratio of apoptosis-related genes Bcl-6/Bax increases, this alteration makes the apoptosis decreased and VSMC proliferated in hypertension rats.
