摘　　要：目的　通过糖尿病大鼠视网膜血管铺片的原位杂交及免疫组化 ,直接证明视网膜血管中碱性成纤维细胞生长因子 (bFGF)蛋白质及mRNA的表达 ,并探讨bFGF在糖尿病视网膜病变 (DR)发展过程中的作用。方法　复制链脲佐菌素 (STZ)糖尿病大鼠动物模型 ,取不同组、不同时期大鼠视网膜的血管进行铺片 ,并在铺片上进行免疫组化及原位杂交 ,检测铺片的血管上bFGF蛋白质及mRNA的表达情况。结果　 ( 1)正常对照组 (M)、血糖恢复组及糖尿病 1个月组(M1 )大鼠视网膜血管的形态及周细胞数均无明显差异 ,bFGF蛋白质及mRNA均无阳性表达。 ( 2 )糖尿病 3个月组 (M3)血管周细胞数减少 (P <0 0 1) ,细胞核可见bFGFmRNA表达 ( 78% ) ,血管壁可见bFGF蛋白表达阳性 ( 5 6% )。 ( 3 )糖尿病5个月组 (M5)血管周细胞数减少 (P <0 0 1)且可见闭塞毛细血管及血栓 ,细胞核bFGFmRNA表达阳性率高于M3为 89% ,血管壁bFGF蛋白质表达也高于M3为 89%。结论　bFGF在STZ大鼠的视网膜血管中有表达 ,且随着病程的延长其表达加强。Objective To identify the expression of basic fibroblast growth factor (bFGF) protein and mRNA in retinal vessel in diabetic rats. Methods The streptozocin (STZ) diabetic rat model was copied,and the retinal vessel from different group rats were digested and prepared.Immunohistochemistry and situ hybridation technique were used to evaluate the function of bFGF during the progress of diabetic retinopathy.The expression of bFGF protein and mRNA were measured. Results (1) The morphology of retinal vasculum and the numbers of the pericytes were insignificantly different in the normal control group,blood glucose recovering group and diabetic M 1 group(diabetes for one month),and bFGF protein and mRNA were not detected in those groups. (2) In the rats with diabetes for three months(M 3),the numbers of the pericytes were decreased (P<0.01),and bFGF mRNA were expressed in nuclear(78%),and expression of bFGF protein was positive in the vascular wall(56%).(3)The numbers of the pericytes were decreased in the rats with diabetes for five months (M 5) (P<0.001),and thrombus and blocking of capillary could be seen.The expression of bFGF were higher than that in M 3 group(89%). Conclusion bFGF can be expressed in retinal vascular cells in STZ rats with a positive correlation with the development of the disease.
