摘　　要：目的 :观察胰岛素在MPP+ 诱导PC12细胞凋亡中的干预作用。方法 :应用MTT法研究细胞活性的改变 ,应用HOECHST3 3 2 5 8染色结合荧光显微镜技术及流式细胞技术检测不同药物对PC12细胞的凋亡诱导作用 ,应用半定量逆转录聚合酶链式反应 (RT PCR)测定胰岛素受体 (insulinreceptor ,IR)mRNA的改变。结果 :①MPP+ 诱导PC12细胞凋亡 ,胰岛素可以抵抗此凋亡作用 ;②以上两种处理 ,均未见到胰岛素受体mRNA的改变 ,推测胰岛素受体的自身磷酸化有改变。结论 :胰岛素可以抵抗MPP+ 诱导的PC12细胞的凋亡。Aim:To study the effect of the insulin against 1-methyl-4-phenylpyridium ion (MPP +)induced apoptosis in pheochromocytoma (PC12) cells.Methods:3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay,HOECHST 33258 staining method,flow cytometric analysis were performed on PC 12 cells,the expression of insulin receptor (IR) mRNA was measured by using RT-PCR.Results:(1)Cell treated with MPP + exhibited various apoptotic features,while cells pretreated with insulin prior to MPP + exposure showed a decrease in the occurrence of apoptotic features.(2)Both MPP + and insulin had no effects on the expression of IRmRNA,the phosphorylation of IR may be a potential mechanism.Conclusion:The insulin may exert a protective effect against MPP +-induced apoptosis in PC12 cells.
