摘 要:目的:探讨水通道蛋白1 (AQP1)对人鼻咽癌细胞迁移及侵袭能力的影响。方法:采用shRNA转染人鼻咽癌细胞5-8F后,用Western blot检测AQP1蛋白的表达量,Transwell实验检测细胞的迁移及侵袭能力。结果:shRNA质粒转染细胞能显著降低人鼻咽癌细胞5-8F中AQP1的蛋白表达水平(P 【0.05)。Transwell实验发现抑制AQP1的表达可降低前列腺癌细胞的迁移侵袭能力(P 【0.05)。结论:AQP1可促进鼻咽癌细胞的迁移和侵袭,可成为鼻咽癌治疗后潜在靶点。Objective: To investigate the effects of AQP1 on nasopharyngeal carcinoma cell migration and in-vasion. Methods: Nasopharyngeal carcinoma cell 5-8F was transfected with short hairpin RNA (shRNA) targeting AQP1. And then, Western blot was used to detect the AQP1 protein expression, and Transwell assay was used to detect cell migration and invasion. Results: The expression of AQP1 was effectively downregulated in nasopharyngeal carcinoma cell by shRNA targeting AQP1. Knockdown of AQP1 inhibited TNBC cell migration and invasion in vitro (P <0.01). Conclusion: Aquaporin 1 can promote the migration and invasionin nasopharyngeal carcinoma cell. Thus, AQP1 may serve as a potential therapeutic target for the treatment of TNBC.
