摘 要:香梨优斑螟是新疆危害林果的重要害虫之一,为研究该害虫某些基因的表达情况,选取β-actin、18s rRNA、UBQ、GAPDH和β-tub等5种候选内参基因为研究对象,利用实时荧光定量PCR技术分析候选内参基因相对表达情况,以Delta Ct、GeNorm、BestKeeper、NormFinder和RefFinder软件或方法评价表达稳定性。结果发现:18s rRNA和UBQ基因在香梨优斑螟不同虫态中稳定性较好,只有BestKeeper软件分析UBQ基因稳定性略好于18s rRNA,其次是TUB和β-Actin,GAPDH表达稳定性最差。Euzophera pyriella is one of the important pests in forestry and fruit trees in Xinjiang.In order to study the expression of some genes in this pest,5 candidate internal reference genes such asβ-actin,18s rRNA,UBQ,GAPDH andβ-tub were selected as the research objects.The relative expression of candidate internal reference genes was analyzed by real-time fluorescent quantitative PCR.The expression stability was evaluated by Delta Ct,GeNorm,BestKeeper,NormFinder and RefFinder soft-ware or methods.The results showed that 18s rRNA and UBQ genes had good stability in insect states of Euzophera pyriella,and only BestKeeper software analyzed UBQ gene stability slightly better than 18s rRNA,followed by TUB andβ-Actin,GAPDH expression stability was the worst.
