类脂褐素模型优化及几种天然抗氧化剂对其体外清除作用研究  被引量:1

摘  要:为确定天然抗氧化剂对脂褐素的清除效果,优化构建了体外类脂褐素模型,通过扫描电子显微镜(SEM)和傅里叶近红外光谱仪(FTIR)研究了天然抗氧化剂对类脂褐素结构的影响。结果表明,类脂褐素体外最优模型为1 mg/mL的牛血清白蛋白(BSA)与2 mmol/L的丙二醛(MDA)在pH7.4的磷酸盐缓冲液中37℃孵育72 h。硫辛酸、抗坏血酸、原花青素、茶多酚、紫薯花青素、槲皮素、芦丁和单宁对类脂褐素的半抑制浓度(IC50)分别为25、0.21、0.18、0.16、0.15、0.13、0.033 mmol/L和0.021 mmol/L。SEM结果显示,蛋白质在生成类脂褐素的过程中发生交联,出现纤维状结构;芦丁和单宁的加入使得类脂褐素晶体结构得到修复,交联程度降低,纤维状结构减少。FTIR分析表明,蛋白质在生成类脂褐素过程中出现了O—H和C—H结构,加入单宁后C—H结构消失,加入芦丁后O—H消失,类脂褐素结构得到改善。芦丁、单宁、槲皮素、紫薯花青素、茶多酚、原花青素和抗坏血酸等对体外类脂褐素均具有较好的清除作用,可作为抗衰老功能食品的良好来源。The lipofuscin model was established and optimized in vitro and the effects of natural antioxidants on the structure of lipofuscin were reserched by scanning electron microscopy(SEM)and fourier near infrared(FTIR)to explore the scavenging effects of natural antioxidants on lipofuscin.The results showed that the optimal model of lipofuscin in vitro was 1 mg/mL bovine serum albumin(BSA)and 2 mmol/L malondialdehyde(MDA)at pH 7.4 phosphate buffer,at 37℃for 72 h.The semi-inhibitory concentrations(IC50)of lipoic acid,ascorbic acid,proanthocyanidins,tea polyphenols,anthocyanins from purple potatoes,quercetin,rutin and tannins against lipofuscin were 25 mmol/L,0.21 mmol/L,0.18 mmol/L,0.16 mmol/L,0.15 mmol/L,0.13 mmol/L,0.033 mmol/L and 0.021 mmol/L,respectively.The SEM results showed that the protein was crosslinked and fibrous in the process of lipofuscin production.With the addition of rutin and tannin,the crystal structures of lipofuscin were repaired,the degrees of cross-linking were decreased,and the fibrous structures were decreased.FTIR analysis showed that O—H and C—H structures appeared in the process of lipofuscin formation.The C—H and O—H structures disappeared,after adding tannin and rutin,respectively and the structures of lipofuscin were improved.Rutin,tannin,quercetin,anthocyanins from purple potatoes,tea polyphenols,proanthocyanidins,ascorbic acid and lipoic acid have good scavenging effects on lipofuscin in vitro,which can be used as good sources of antiaging functional products.
