摘 要:目的探讨柚皮素(NAR)对APP/PS1转基因小鼠淀粉样蛋白沉积和胶质细胞活化的抑制作用。方法利用APP/PS1转基因小鼠观察NAR对AD小鼠在水迷宫实验中记忆损伤的改善作用;硫磺素-S染色法检测淀粉样斑块沉积;免疫荧光法检测SYN-1水平;蛋白免疫印迹法测定小鼠大脑SYN-1、PSD-95和GAP-43含量;免疫荧光法检测GFAP和Iba-1水平。结果水迷宫实验结果显示NAR可明显降低转基因小鼠逃避潜伏期并增加穿越平台所在象限次数。硫磺素-S染色结果显示,NAR可明显降低小鼠皮层及海马淀粉样斑块沉积。蛋白免疫印迹实验显示,NAR可增加SYN-1、PSD-95和GAP-43表达水平。免疫荧光实验显示,NAR可明显降低APP/PS1转基因小鼠皮层和海马GFAP和Iba-1荧光强度。结论NAR可通过减少皮层和海马中淀粉样斑块沉积并抑制胶质细胞活化改善APP/PS1转基因小鼠记忆功能受损。Objective To investigate the inhibitory effect of naringenin on amyloid protein deposition and glial cell activation in APP/PS1 double transgenic mice.Methods APP/PS1 transgenic mice were used to observe the effect of NAR on memory impairment of AD mice in a water maze experiment;Thioflavin-S staining was used to detect amyloid plaque deposition;immunofluorescence was used to detect SYN-1 level;by western blot,the levels of SYN-1,PSD-95 and GAP-43 in the brain of mice were measured;the levels of GFAP and Iba-1 were detected by immunofluorescence.Results The results of the water maze experiment showed that NAR could significantly reduce the escape latency of transgenic mice and increase the number of times the mice crossed the quadrant with the platform.Thioflavin-S staining showed that NAR significantly reduced amyloid plaque deposition in the cortex and hippocampus of mice.Western blot showed that NAR could increase the expression levels of SYN-1,PSD-95 and GAP-43.Immunofluorescence assay showed that NAR could significantly reduce the fluorescence intensity of GFAP and Iba-1 in cortex and hippocampus of APP/PS1 transgenic mice.Conclusion NAR improves memory impairment in APP/PS1 transgenic mice by reducing amyloid plaque deposition in cortex and hippocampus and inhibiting glial cell activation.
