摘　　要：采用单因子和正交试验对毕赤酵母重组菌株产生人凝集素样氧化型低密度脂蛋白受体-1(rhLOX-1)的发酵过程进行了优化,确定了最佳发酵条件:0.8%甲醇,1.8%酪蛋白水解物,初始pH 7.0,3×接种量。在此培养条件下,rhLOX-1在毕赤酵母重组菌株中的表达量可超过100 mg/L,比对照提高了1倍以上。本研究为大规模发酵制备rhLOX-1蛋白奠定了基础,更好地满足了LOX-1的理论研究及新药筛选的需要。The technology of liquid fermentation for producing lectin-like oxidized low-density lipoprotein receptor-1(LOX-1) in Pichia pastoris was studied by using single-factor and orthogonal test.The results showed that the optimal culture conditions were as follows:0.8% methnol,1.8% casamino acids,intial pH 7.0 and three times of basal inoculation volume.Under the above culture conditions,the expression level of LOX-1 in recombinant strains was above 100mg/L,which was more than twice of the control.The study has laid a foundation for the large-scale preparation of LOX-1 to meet the needs of theoretical research of LOX-1 and screening of new medicines.
