摘 要:目的探索中国延边朝鲜族、汉族人群的PGC-1基因Ser74Leu、IVS2+52C>A多态与2型糖尿病(T2DM)的相关性。方法采用聚合酶链反应-单链构象多态性技术(PCR-SSCP)确定PGC-1基因的第2外显子及第2内含子的Ser74Leu、IVS2+52C>A多态位点的基因型,比较等位基因频率及基因型频率在民族之间差异。结果1.朝鲜族和汉族的T2DM、NGT组间的PGC-1基因Ser74Leu(C>A)、IVS2+52(C>A)两个等位基因三种单体型的频率分布比较未见差异(P=0.178,P=0.671);2.T2DM组中,朝鲜族表型为CC-CA/CC-AA的腰臀比(WHR)水平较表型为CC-CC的高(P=0.030),汉族表型为CC-CA/CC-AA的空腹血糖(FPG)水平高于表型为CC-CC(P=0.005)。结论PGC-1基因可能是中国延边朝鲜族和汉族2型糖尿病的发病的易感基因。Objective Explore the association between two Single nucleotide polymorphisms, (Ser74Leu、IVS2+52C>A) of Peroxisome Proliferator-Activated Receptor-γ Coactivator 1(PGC-1) with type 2 diabetes in Yanbian koreans humans of China. Methods Using PCR-SSCP to determine the genotype of Ser74Leu, IVS2+52C>A, Two SNPs were chosen from the second exon and the second intron of PGC-1. Results 1. There were no significant difference in Haplotype frequencies of Ser74Leu-IVS2+52C>A between T2DM and NGT in Korean-China humans and The Han Nationality (P=0.178, P=0.671); 2.In T2DM group, the level of WHR with the CC-CA/CC-AA was higher than the one with CC-CC in Korean-China humans (P=0.030); the level of FPG with the CC-CA/CC-AA was higher than with CC-CC in the Han Nationality(P=0.005). Conclusion PGC-1 gene might contribute to the high risk of type 2 diabetes in the Korean-China studied here.
