摘  要:目的:以鱼藤酮诱导PC12细胞损伤作为PD细胞模型,观察5-脂氧酶(5-lipoxygenase,5-LOX)是否参与细胞损伤。方法:在鱼藤酮处理后不同时间,以MTT方法检测PC12细胞活性变化,以免疫细胞化学方法观察5-LOX核膜移位情况;并观察5-LOX抑制剂齐留通的作用。结果:鱼藤酮(0.3~30μmol/L)可诱导PC12细胞损伤;齐留通(3~100μmol/L)减轻鱼藤酮诱导的细胞损伤。鱼藤酮还时间和浓度依赖性引起5-LOX核膜移位,齐留通(1~30μmo/L)则能抑制5-LOX核膜移位。结论:5-LOX参与鱼藤酮诱导的PC12细胞损伤,5-LOX抑制剂齐留通可抑制5-LOX激活,减轻细胞损伤。Objective: To determine whether 5-lipoxygenase(5-LOX)is involved in rotenone-induced injury in PC12 cells,which is a cell model of Parkinson disease.Methods: After rotenone treatment for various durations,cell viability was determined by colorimetric MTT reduction assay,and 5-LOX translocation was detected by immunocytochemistry.The effect of 5-LOX inhibitor zileuton was also investigated.Results: Rotenone(0.3-30 μmol/L) induced PC12 cell injury,and zileuton(3-100 μmol/L) attenuated this injury.Rotenone also time-and concentration-dependently induced 5-LOX translocation into the nuclear envelope,and zileuton(1-30 μmo/L) significantly inhibited rotenone-induced 5-LOX translocation.Conclusion: 5-LOX is involved in rotenone-induced injury in PC12 cells,and 5-LOX inhibitor zileuton can reduce rotenone-induced 5-LOX activation and cell injury.
