摘 要:目的观察依达拉奉(edaravone)对小鼠视网膜光损伤(retinal photodamage,RP)的保护作用,并探讨其抗氧化功能。方法 48只成年雄性BALB/C小鼠随机分为正常组(n=12)、光损伤组(n=12)、依达拉奉组(n=12)和生理盐水组(n=12)。采用(8 000±300)lux白色光源照射4h建立小鼠视网膜光损伤模型;应用光镜观察各组小鼠视网膜形态学变化;检测光照后小鼠视网膜组织抑制羟基自由基(HO.)的能力以及丙二醛(MDA)和超氧化物歧化酶(SOD)含量。结果光镜检查显示,依达拉奉组视网膜损伤程度相对于光损伤组和生理盐水组明显减轻(P<0.05);氧化指标检测显示相对于光损伤组和生理盐水组,依达拉奉组抑制HO.的能力显著提高(P<0.05),视网膜组织MDA含量明显减少(P<0.05),SOD含量显著增多(P<0.05)。结论氧化应激在RP的发生中起着关键作用,依达拉奉可以通过抗氧化功能对RP起到保护作用。Objective To observe the protective effect of edaravone on retinal photodamage and explore its antioxidative function. Methods Totally 48 adult BALB/C male mice were randomly divided into normal group(n=12),photodamage group(n=12),edaravone group(n=12) and saline group(n=12).Mice were exposed to(8 000±300)lux white light for 4 hours to establish retinal photodamage models.We used light microscope to test the retinal morphologic changes,and then tested the retina's ability to inhibit hydroxyradicals and the levels of malondialdehyde(MDA) and superoxide dismutase(SOD). Results Light microscopy test showed that retinal damage was less severe in edaravone group than in photodamage group and saline group(P<0.05).The test of oxidation indicator showed that edaravone group had significantly lower HO· and MDA levels but an increased SOD level compared with photodamage group and saline group(all P<0.05). Conclusion Oxidative stress plays a pivotal role in retinal photodamage,and edaravone can protect retina from photodamage through its antioxidative function.
