响应面分析法对碱法提取酒糟蛋白工艺的优化

摘  要:以酒糟为原料,利用碱水解法提取蛋白质,并在单因素试验的基础上,选取料液比、反应温度、反应时间作为影响蛋白提取率的因素。根据 Box-Behnken 中心组合试验设计原理,采用三因素三水平响应面分析法对酒糟蛋白提取工艺进行优化研究,确定了碱法提取酒糟蛋白质时的最佳工艺参数为液料比为 29.75:1,反应温度为 90.51 ℃,反应时间 105 min。在此条件下酒糟蛋白的提取率为 10.6077 %。Alkaline hydrolysis method was used to extract protein from distiller's grains. The main variables influencing the extraction included the ratio of distiller's grains and extracting solvent, reacting time, and reacting temperature. According to the principles of Box-Behnken central composite design, three factors and three levels response surface method was employed to optimize the extraction conditions and the optimum technical parameters were summed up as follows: the ratio of extracting solvent and distiller's grains was 29.75 ∶1, reacting temperature was at 90.51 ℃, and reacting time was 105 min. Under the above conditions, the extraction efficiency of protein was 10.6077 %.
