摘　　要：对荧光法测定磷酯酶C对血小板细胞质游离钙离子浓度作用的实验条件进行了研究.结果表明,在本实验条件下,Fura 2／AM合适的浓度为4μmol／L;最适pH为7.4;血小板的浓度在1×108～3×108个／L;保护剂ASA及激活剂ADP的浓度分别为0.937mmol／L和20mmol／L;波长选用350nm和380nm;背景值对实验结果没有影响.The experimental conditions to detect human platelet cytoplasmic free calcium were studied in human and rat by Fura-2/AM fluorescence technique. The results suggest that the appropriate concentration is 4μ mol / L and the appropriate pH is 7.4. The hematoblastic concentration is 1～3×10~ 11 /L. The concentration of ASA and ADP is respectively 0.937 m mol / L and 20mmol / L. The wavelengh is respectively 350nm and 380nm. The background has little effect on the detection.
