摘　　要：采用层析法分离生物活性多肽,实验选择胃蛋白酶水解样品作为研究模型,重点考察了洗脱流速、柱层高度、吸光度、洗脱缓冲液类型等因素对分离效果的影响,通过分析分离图谱,最终确定了优化的凝胶层析分离条件,具体如下:采用SephadexG15凝胶、pH 7.1的Tris-HCl缓冲液、紫外检测波长为280 nm、床层高度为57.5 cm。Sephadex gel-filtration was used to separate bioactive peptides;bioactive peptides that are released from proteins by enzymatic proteolysis by pepsin were chosen as model peptides. Various parameters including the luminosity, buffer, the column level altitude were investigated to optimize the separation conditions. The optimized conditions are as follows:Sephadex G-15 column (1.7 cm×57.5 cm), pH 7.1 Tris-HCl, 280 nm.
