摘 要:目的探讨大蒜素与奥沙利铂独用和联用对大肠癌细胞HCT-8生长的影响及可能的机制。方法体外培养HCT-8细胞,分成4组,大蒜素组用终浓度为9μg/mL的大蒜素处理,奥沙利铂组用终浓度为5μg/mL的奥沙利铂处理,联用组同时用终浓度为9μg/mL的大蒜素和5μg/mL的奥沙利铂处理,对照组用终浓度为0.5%的DMSO处理。处理72 h后采用MTT法检测各处理组HCT-8细胞的增殖情况,流式细胞仪检测各组细胞周期及细胞凋亡情况,免疫组化法检测各组caspase-3表达情况。结果联用组HCT-8细胞的增殖抑制率明显大于大蒜素组和奥沙利铂组,差异有统计学意义(P<0.01)。细胞周期检测结果显示:联用组G0/Gl期细胞数明显少于对照组,G2/M期细胞数明显多于对照组,差异有统计学意义(P<0.01);联用组凋亡率明显高于单用组,差异有统计学意义(P<0.01)。联用组caspase-3的阳性表达率明显高于单用组,差异有统计学意义(P<0.01)。结论大蒜素与奥沙利铂合用对HCT-8细胞的生长具有协同抑制作用,其作用机制可能与抑制细胞增殖、阻滞细胞周期及促进细胞凋亡有关。Objective To investigate the effect of allicin and oxaliplatin on colorectal cancer HCT-8 cell growth and its mechanism.Methods HCT-8 cell were cultured in vitro and divided into four groups,allicin group HCT-8 cell were treated with final concentration 9 μg /mL allicin,oxaliplatin group were treated with final concentration 5 μg /mL oxaliplatin,combination group were treated with both allicin and oxaliplatin,with final concentration 9 μg /mL and 5 μg /mL,control group were treated with final concentration 5% DMSO. After 72 hours,the proliferation of HCT-8 cell in each group were detected by MTT method,cell cycle and apoptosis were examined by flow cytometry,expression of Caspase-3 were tested by immunohistochemical method. Results Inhibition rate of HCT-8 cell proliferation in combination group was significantly higher than that of garlicin group and oxaliplatin group(P < 0. 01). Cell cycle test results showed that cell number of G0 /Gl phase in combination group was significantly less than control group,the cell number of G2 /M phase was higher than control group(P < 0. 01); the apoptosis rate of combination group was significantly higher than two single groups(P < 0. 01); the positive expression rate of Caspase-3 group was significantly than the two single group(P < 0. 01). Conclusion Allicin and oxaliplatin have synergistic inhibitory effect on HCT-8 cell growth,its mechanism may be related to the inhibition of cell proliferation,cell cycle arrest and cell apoptosis.
