摘 要:【目的】研究糯玉米自交系中游离氨基酸的组成及含量,对糯玉米种质资源的营养品质进行评价。【方法】采用邻苯二甲醛(OPA)柱前衍生反相高效液相色谱法(RP-HPLC)测定了60份糯玉米自交系籽粒的游离氨基酸组成及含量,并采用主成分分析法进行综合评价。【结果】从60份糯玉米自交系中共检测出16种氨基酸,最少的检测出14种氨基酸,其中JN57的氨基酸总量最高(25.165mg/g),JN21的氨基酸总量最低(6.403mg/g),平均为12.236mg/g,含量高于平均值的有25份。人体必需氨基酸(EAA)含量在2.47~13.97mg/g,占游离氨基酸总量的29.43%~75.52%。对60份糯玉米自交系所含的16种氨基酸进行主成分分析及综合评价,得分值在前3位的依次为JN57、JN59和JN55。【结论】60份糯玉米自交系的游离氨基酸含量具有明显的遗传多样性,其中JN57、JN59及JN55的品质较优,JN21的品质最差。【Objective】This study aimed to analyze the components and contents of free amino acids in waxy maize inbred lines and evaluate the nutritional quality of germplasm resources.【Method】An effective reversed-phase high performance liquid chromatography(RP-HPLC)after pre-column derivatization with o-phthaladehyde(OPA)was used to determine the components and contents of free amino acids in 60 waxy maize inbred lines and principal component analysis was conducted for comprehensive evaluation.【Result】A total of 16(with the minimum of 14)free amino acids were detected from the 60 inbred lines.JN57 had the highest total content of 25.165mg/g while JN21 had the lowest of 6.403mg/g.The average content was 12.236mg/g and 25 samples exceeded the average.The contents of essential amino acids(EAA)varied from 2.47 to 13.97mg/g,accounting for 29.43%-75.52% of total free amino acids.Principal component analysis showed that first three inbred lines with the highest scores were JN57,JN59,and JN55.【Conclusion】The contents of free amino acids in 60 waxy maize inbred lines showed clear genetic diversity.JN57,JN59 and JN55had the best quality while JN21 had the worst.
