摘 要:目的 探讨针刺体壁对丝光绿蝇抗菌物质的诱导表达。并研究丝光绿蝇免疫血淋巴的性质。方法 丝光绿蝇3龄幼虫针刺体壁处理后,分别于第0、12、24、36、48、60、72、84h提取血淋巴,以金黄葡萄球菌为指示菌,采用平板滤纸片法作抑菌试验,用于显示丝光绿蝇抗菌物质的诱导表达,以抑菌圈直径表示抑菌活力的大小。提取诱导后48h的血淋巴,设置3μl、5μl、7μl不同的血淋巴加样量实验组,记录不同加样量对金黄葡萄球菌的的抑菌效果;设置不同稀释浓度,记录不同浓度的血淋巴对金黄葡萄球菌的抑菌活力。诱导后48h的血淋巴置于冰箱保存后,设置36℃和100℃不同解冻温度,观察不同温度解冻的血淋巴对金黄葡萄球菌和大肠杆菌的抑菌效果。诱导后48h的血淋巴,100℃水浴3min,观察其对金黄葡萄球菌的抑菌活力。提取诱导后48h的血淋巴,观察其对大肠杆菌、金黄葡萄球菌、白色葡萄球菌、变形杆菌、伤寒杆菌、副伤寒杆菌、枯草杆菌、溶壁微球菌等8种细菌的抑菌效果。结果 丝光绿蝇幼虫针刺体壁处理后,对金黄葡萄球菌的抑菌活力高峰出现在处理后48h。平板滤纸片法抑菌试验中,以加7μl血淋巴的抑菌效果最佳。诱导后48h的血淋巴置于冰箱保存后,36℃和100℃不同温度解冻,对金黄葡萄球菌和大肠杆菌均有抑菌效果,36℃解冻效果好于100℃。?To investigate the kinetics of the inducible antibacterial activities of the hemolymph in larvae of blowfly Lucilia ser-icata, the hemolymph was collected at different times after induction by pricking and the antibacterial activities of the hemolymph induced were determined by the paper disc method of bacterial growth inhibition, using Staphylococcus aureus as the indicator organism. It was found that the peak of antibacterial activity of the hemolymph in larvae against S. aureus appeared at 48 hours after induction with a better result in 7 μl dosage of hemolymph. The effect of thawing at 36℃ was better than that at 100℃ . Definite antibacterial activities of hemolymph against Escherichia coli, Staphylococcus aureus, Staphlococcus albus, Proteus vulgaris, Salmonella typhi, Salmonella paratyphi, Bacillus subtilis and Micrococcus lysodeikticuswere also demonstrated. This hemolymph also possessed antibacterial property against S. aureus and E. coli after incubation at 100℃for 3 min and against S. aureus after dilution for 64 times. It concludes that the hemolymph in larvae induced by pricking possess a wide range of antibacterial spectrum with a definite thermosta-bility.
