摘 要:从南汇县黄路乡一草堆下的土壤中分离到一株产耐热蛋白酶的链霉菌.该酶的最适反应温度为70℃.在60℃时稳定,当80℃时处理10min,尚余84%活性.在Ca^(2+)存在的条件下,酶的热稳定性有显著提高.酶反应的最适pH为8.5,在pH5~9之间稳定,因此属于微碱性蛋白酶.通过抑制剂试验,该酶基本上被PMSF(苯甲基磺酰化合物)抑制,但不被金属螯合剂(EDTA,双吡啶)抑制.金属离子Cu^(2+),Zn^(2+),Fe^(2+)和Co^(2+)对酶有抑制作用,Ca^(2+)对酶有激活作用.A thermophilic Streptomyces producing extracellular proteinase in high was isolated from soil. The protease has an optimum reaction temperature of 70℃ and was stable at 60℃. It retains 84% activity even after treatment of 80℃ for 10 min. Ca^(2+) enhanced the heat stability of the enzyme obviously. The optimum pH for casein hydrolysis was 8.5. The enzyme was stable over the pH range from 5 to 9. The enzyme was inactivited after treatment with PMSF, but not inactived by EDTA and a-a′Bipyridin. The cation such as Cu^(2+), Zn^(2+), Fe^(2+) and inhibited the enzyme activity while Ca^(2+) trends to stimulate activity.
