摘 要:为了研究对EBV LMP2基因按照真核密码子偏好进行密码子优化后 ,对其在真核细胞蛋白表达水平以及引发细胞免疫能力的影响 ,我们对EBV LMP2基因进行了完全改造 ,并构建了改造前后的Psectag/LMP2重组质粒 ,通过瞬时转染COS 7细胞检测其蛋白表达并进行了小鼠的DNA疫苗实验。结果表明 ,改造后的LMP2基因在真核细胞中的表达未见大幅提高 ,同时引发细胞免疫的能力也没有明显上升。结果提示 ,我们单纯按照密码子的偏好来改造基因并不一定能使蛋白的表达量和引发免疫反应的能力提高。To observe the influence on the protein expression level in mammalian cell and the ability to elicit cell immunity of EBV-LMP2 gene after its codons were optimized according to mammalian codon bias,the gene of EBV-LMP2 was fully modified,and the plasmids Psectag/LMP2 containing two genes were constructed. The protein expression level was detecte by transient transfection in COS-7 cell,at the same time the DNA vaccines were inoculated into mice. The results showed that the protein expression level was not elevated markedly,and the ability to elicit cell immunity was not enhanced too. This result suggests that the method to modify gene simply according to codons bias can not always elevate the protein expression level and improve the immunogenic character.
