信号肽重组HBD2在人肺癌细胞中表达的初步研究

摘　　要：目的　构建高表达人源化β 防御素 2 (HBD2 )表达体系。 方法　构建人pLNCX2 CEA信号肽 HBD2 (成熟肽 )逆转录表达载体 ,DOTAP转染逆转录包装细胞 ,获得含融合基因的侵袭性逆转录病毒 ,再转染人肺癌A5 4 9细胞 ,用免疫印迹法鉴定转化细胞中HBD2蛋白的表达。结果　成功构建了表达HBD2成熟肽的重组信号肽HBD2的人肺癌表达体系 ,经WesternBlot检测 ,证实有目的基因表达。Objective To construct high expression system of human β defensin 2(HBD2) in human lung cancer cell line A549.Methods A recombinant retrovirus expression vector pLNCX2 CEA signal peptide HBD2(mature peptide sequence) was constructed,and the retrovirus vector was transfected into PT67 cell by DOTAP.After screening and amplification of single clone cell by G418,the virus was used to infect A549 cell.The A549 cells were also been screened by G418,and cell clone resistant to G418 was got.Expression of HBD2 was detected by western blotting analysis.Results A recombinant retrovirus expression vector pLNCX2 CEA signal peptide HBD2(mature peptide sequence) was constructed successfully,and high level of HBD2 expression was obtained in A549 cell transfected with the expression vector.Conclusion With gene engineering technology,human lung cancer cell line A549 can be used for high expression of HBD2.
