白血病细胞核DNA空间构象的荧光研究

摘  要:DNA的空间构象与传递信息功能的研究已开始受到重视。DNA的空间构象,荧光物质菲啶溴红(EtBr)插入DNA双链区的量,形成的EtBr-DNA镶嵌结构中EtBr的荧光量子率的增加程度均不同。本文根据上述原理,应用EtBr荧光滴定法测定比较了正常人白细胞核DNA与白血病细胞核DNA的结果。通过Scatchard公式计算并绘出曲线证明正常白细胞与白血病细胞的核DNA构象是不同的,这可能是它们表达功能不同的有力依据。It has been begun to notice that the study of relation between the steric conformation and function of DNA. If the steric conformation of DNA was different, the content of fluorescence substance EtBr which intercalated DNA double helix structure was also different. According to above mechanism, we used EtBr fluorescence method to determine the steric conformation of normal white blood cell nuclear DNA and leukemic cell nuclear DNA. The Scatchard curve was made through formula calculation. The result demonstrated that the steric conformation of cellular nuclear DNA between normal white blood cell and leukemic cell was different, and this may be the important base that they expressed different function.
