PTEN基因蛋白在乳腺癌组织中的表达及其临床意义

摘　　要：目的 :探讨抑癌基因PTEN蛋白的表达水平对判断乳腺癌生物学行为和预后的临床意义。方法 :采用免疫组织化学S P法 ,检测 42例石蜡包埋乳腺癌组织和 40例乳腺小叶增生组织中PTEN蛋白的表达水平。结果 :PTEN蛋白在乳腺癌组织中的高表达率比乳腺小叶增生低 (P <0 0 1)。PTEN基因蛋白表达与肿瘤大小、组织学分级、临床分期和肿瘤复发无相关性 ,<45岁患者PTEN表达比 >45岁偏低 (P <0 0 5 ) ;PTEN高表达与腋淋巴结转移情况有显著负相关性 (P <0 0 1) ;与ER受体呈正相关性。PTEN基因蛋白高表达与远处转移有负相关性 ;与 5年生存率呈正相关性 (P <0 0 1)。结论 :PTEN蛋白表达异常与乳腺癌发生、发展相关 。Objective:To observe the clinical value of expression of PTEN gene protein for predicting biological behaviors and prognosis of breast cancer. Method: Immunohistochemistry(S-P method)was used to detect the expression of PTEN in forty-two cases of paraffin-embedded human breast cancer tissues, and forty cases of paraffin-embedded hyperplasia of mammary glands tissues. Results: The high expression rate of PTEN gene protein was less in breast cancer than in hyperplasia of mammary glands(P<0.01). The expression of PTEN gene protein was not related with the tumor size, histological grade, clinical stage and postoperative recurrence; but the high expression rate was higher in the elder patients(more than 45 years)than in younger(less than 45 years). The high expression of PTEN was significantly inversely correlated with axillary lymph node metastasis(P<0.01)and it was a positive relationship with estrogen receptor(P<0.05). The high expression of PTEN in breast cancer was negatively correlated with distant metastasis(P<0.05)and was an positive relationship with the survival rate of 5-year(P<0.01).Conclusion:The abnormal expression of PTEN gene protein plays an important role in the development of primary breast carcinoma. It may be a predictor for biological behaviors and prognosis of breast cancer.
