摘 要:目的 :利用大鼠福尔马林致痛模型 ,探索精氨酸加压素 (AVP)的镇痛作用以及其在乙酰水杨酸 (ASA)中枢镇痛中的作用。方法 :实验大鼠 (2 4只 )随机分为 3组 ,每组 8只 ,N组足背注射生理盐水 ,F组足背注射 5 %福尔马林 ,A组足背注射 5 %福尔马林后立即腹腔注射ASA。采用痛行为评分法观测大鼠的痛反应 1小时后 ,进行免疫组织化学SABC法 ,观察AVP的免疫反应性 (AVP -ir) ,并进行图像半定量分析。结果 :N组无明显疼痛反应 ,F组大鼠给药后迅速出现疼痛反应 (缩腿 ,舔爪 ) (P <0 .0 1) ,与F组比较 ,A组疼痛反应明显减轻 (P <0 .0 1) ;F组大鼠下丘脑视上核 (SON)内AVP -ir较N组增强 ,室旁核 (PVN)与视交叉上核 (SCN)内AVP -ir较N组均减弱 ;给予ASA后 ,SON与PVN内AVP -ir较F组均减弱 ,SCN内AVP -ir较F组有所增强。结论 :下丘脑SON、PVN、SCN神经元分泌的AVP可参与中枢镇痛 。Objective:The experimental aim is to use rat pain model induced by formalin (F) to explore the analgesic pain effect of arginine vasopressin (AVP) and the center analgesia effect of AVP on acetylsalicylic acid (ASA). Methods:24 Wistar rats were randomly divided into normal saline (N) group, formalin (F) group, ASA (A) group. The pain praxiology score and Immunohistochemistry SABC method were used, then image semi-quantity method was used to analysize AVP-immunocreative (AVP-ir).Results:The F group rats showed spontaneous pain-related responses (flinching or shaking and licking or biting injected paws) after being injected formalin into one hindpaw, and the spontaneous pain-related responses of A group were significantly relieved; The immunocreative of AVP (AVP-ir) in supraoptic nucleus(SON) was significantly greater than those of N group,the AVP-ir in paraventricular nucleus (PVN) and suprachiasmatic nucleus(SCN) were significantly lower than those of N group;After given ASA,the AVP-ir in SON and PVN were significantly lower than those of F group,but the AVP-ir in SCN was significantly greater than those of F group.Conclusion:The AVP released from SON?PVN and SCN all participate in center pain modulation, ASA can significantly reinforce the effect by regulating synthesis and release of AVP.
