摘 要:目的:观察转入正常PTEN基因的胶质瘤细胞体外粘附力改变,探索PTEN基因对胶质瘤细胞影响方式。方法:构建携带正常PTEN基因的pcDNA3.1/Hygro(-)真核质粒载体,并用之转染PTEN失活的U251细胞系,采用层粘连蛋白、纤粘连蛋白粘附模型,观察转染前后的细胞的粘附力的改变。结果:转染PTEN基因可以明显抑制U251细胞的粘附力。Objective: To study the effects of wild-type PTEN on the adhesion of gliomas cell.Method: Construct a pcD-NA3.1/Hygro ( - ) recombinant plasmid containing PTEN gene. U251 cells which express no PTEN were transfected with wild-type PTEN plasmid vector and stable transfected clones were selected. Then, the different adhesion ability of three cells were observed, including the transfected cells, untransfected cells and the cells transfected with empty vector. Results: The abilities of adhesion of the transfected cells were obviously inhibited. Conclusion: Wild-type PTEN could have a positive impact on the adhesion of gliomas cell.
