摘 要:目的:探讨高糖对人肾小球系膜细胞活性氧、Ⅳ型胶原合成的影响及其茶多酚的干预作用。方法:系膜细胞在高糖(35mmol/LD-糖)条件下加入或不加入茶多酚(20μg/ml)培养至36h,运用化学比色法、放射免疫法检测高糖作用后培养系膜细胞上清液中活性氧(SOD和MDA)、Ⅳ型胶原的含量以及加入茶多酚后其含量的改变;应用免疫细胞化学法检测高糖作用后系膜细胞内Ⅳ型胶原的表达以及加入茶多酚后其表达的改变。结果:高糖使系膜细胞上清液中SOD含量减少,MDA含量增加,使培养的系膜细胞上清液中及细胞内Ⅳ型胶原含量增加;茶多酚可提高上清液中SOD活性,降低MDA含量,抑制系膜细胞合成和分泌Ⅳ型胶原。结论:高糖致人肾小球系膜细胞活性氧产生增加,促进Ⅳ型胶原合成和分泌,茶多酚可有效干预高糖的作用。Objective: To investigate the effect of high glucose on the production of reactive oxygen species and type Ⅳ collagen in human mesangial cells and the intervention of tea polyphenols(TPs). Methods: The cells were cultured in high glucose with or without TPs up to 36 h. The supernatants were collected and the activities of SOD and the level of MDA were detected by spectrophotometry. The content of type Ⅳ collagen in the media was detected by radioimmunoassay. Immunocytochemistry was used to detect the expression of type Ⅳ collagen within the cells. Results: SOD was lowered, MDA increased, while type Ⅳ collagen increased in the high glucose media without TPs. However, SOD increased, MDA was lowered , type Ⅳ collagen was lowered with TPs compared with that only with high glucose. Conclusion: High glucose contributes to the generation of reactive oxygen species, an increase in type Ⅳ collagen produced by human cells and TPs can better control high glucose-induced reactive oxygen species, type Ⅳ collagen.
