短暂性前脑缺血后大鼠海马各区域bcl-2家族表达变化的研究

摘　　要：目的观察大鼠前脑缺血再灌流后海马各区域bcl-2家族的表达变化规律.方法免疫组织学和图象处理技术.结果 (1)前脑缺血再灌流后6h、12h、24h,bax在CA1区的表达明显增加(49.1±8.0～59.2±6.3 vs 50.1±4.0～72.3±8.9,P ＜0.05);再灌流后12h,Bax在CA3区的表达略有增加(P ＜0.05);而Bax在齿状回的表达始终无明显变化.(2)在前脑缺血再灌流后12h、24h,bcl-2及bcl-xl在海马三个亚区的表达都明显增加(bcl-2:72.7±9.6～95.6±6.4;bcl-xl:67.7±6.4～88.1±9.0,P ＜0.05). 结论 CA1区在缺血性脑损伤的早期即出现Bax的表达增加,这个变化可能是CA1区较其他亚区更易产生迟发性神经元缺失的重要原因之一.Objective To observe the changes in protein expression of bcl-2 family in rat hippocampal subfields following transient forebrain ischemia.Methods Immunohistochemistry and computer-assisted analysis techniques were used. Results(1) The increase in Bax expression was seen at 6h, 12h and 24h after ischemia in the CA1 subfield(P<0.05). In the CA3 subfield ,the increase in Bax expression was only seen at 12h after ischemia(P<0.05). No change of Bax expression was found in the dentate gyrus after ischemia. (2) In all hippocampal subfields , the increase in bcl-2 and bcl-xl expression was found at 12h and 24h after ischemia(P< 0.05).Conclusion The results indicate that the increase in Bax protein expression at early after ischemia might account for the ischemia-vulnerable nature of the CA1 subfield partly.
