摘  要:目前所采用的氨基酸分析法,在柱前或柱后衍生化后,大多数都用价格昂贵的荧光检测器进行测定。近年来已有人采用一些新的衍生化试剂和方法,改用紫外吸收直接测定,取得了很大成功。我们采用1-氟-2,4-二硝基苯(FDNB)A new method for separation and quantification of primary and secondary amino acids by pre column derivatization with 1-fluoro-2, 4-dinitroben-zene and reversed-phase HPLC is described. The products 01 derivatization are more stable in solution than those with traditional methods. A simple linear gradient elution with acetate buffer and ace-tonitrile solution allows the separation of more than 16 amino acids in 40 min at room temperature. The method has been successfully applied to the determination of amino acid composition of pig plasmin. It has good reproducibility and high recovery.
